Research Description
My laboratory is performing studies on two genes that regulate fasting blood glucose levels and thereby contribute to the risk of diabetes and cancer in humans. Some of our current studies on these two genes are as follows:
1. G6PC1, also known as G6Pase, is mainly expressed in liver. It encodes a glucose-6-phosphatase catalytic subunit, which catalyzes the terminal step in glycogenolysis and gluconeogenesis. Increased expression of G6PC1 contributes to elevated fasting blood glucose levels and the risk of developing type 2 diabetes and cancer. Increased G6PC1 expression also contributes to the increased liver glucose production that is characteristic of individuals who have developed either type 1 or type 2 diabetes. This increased glucose production contributes to the complications associated with these diseases, such as retinopathy and kidney failure. Our studies are focused on the molecular signaling pathways that mediate the inhibition of G6PC1 enzyme activity by insulin. These experiments involve the use of tissue culture cells. We are also collaborating with Derek Claxton and Hassane Mchaourab to elucidate the structure of G6PC1.

G6PC2 controls the set point for glucose-stimulated insulin secretion but, just like G6PC1, increased expression of G6PC2 contributes to elevated fasting blood glucose levels and the risk of developing type 2 diabetes and cancer. A major goal of our studies is to elucidate the biological function of G6PC2. We are currently performing studies on G6pc2 knockout mice and islets to address this question. We are also performing molecular studies to identify key amino acids that are important for G6PC2 enzyme activity and using BioVU to link mutations in the G6PC2 gene with disease in humans.
In previous years, rotation projects have been designed to enable students to learn PCR and tissue culture, techniques that are used in multiple laboratories at Vanderbilt. However, students that are already familiar with these techniques would have the opportunity to participate in experiments on knockout mice and mouse islets as well as recently funded studies designed to identify G6PC1/G6PC2 inhibitors.

My laboratory is interested in the transcriptional regulation of genes whose altered expression may contribute to the pathophysiology of diabetes. We are currently working on two such genes as follows:
1. Glucose-6-Phosphatase. Glucose-6-phosphatase (G6Pase) catalyzes the terminal step in glycogenolysis and gluconeogenesis. Increased expression of G6Pase contributes to the increased hepatic glucose production characteristic of both type I and type II diabetes. Our studies mainly focus on the molecular mechanisms that mediate the regulation of G6Pase gene transcription by hormones, especially insulin.
2. IGRP. We have cloned a novel gene that encodes an islet-specific glucose-6-phosphatase catalytic subunit-related protein (IGRP). The goal of the on-going experiments in this project is the elucidation of the molecular basis for the islet-specific expression of the IGRP gene. These experiments involve the use of both tissue culture cells and transgenic mice. The data suggest that the regulation of IGRP gene expression is determined, in part, by novel factors. As such, the identification of these novel factors will potentially aid other investigators who are attempting to understand the process whereby islet stem cells differentiate towards that of a beta cell lineage. In addition, although its biological function is unknown, IGRP has recently been identified as a major autoantigen in type 1 diabetes. Genetic data suggests that suppression of IGRP gene expression might prevent the development of type 1 diabetes in children.

My laboratory is interested in the transcriptional regulation of genes whose altered expression may contribute to the pathophysiology of diabetes. We are currently working on two such genes as follows:
1. Glucose-6-Phosphatase. Glucose-6-phosphatase (G6Pase) catalyzes the terminal step in glycogenolysis and gluconeogenesis. Increased expression of G6Pase contributes to the increased hepatic glucose production characteristic of both type I and type II diabetes. Our studies mainly focus on the molecular mechanisms that mediate the regulation of G6Pase gene transcription by hormones, especially insulin.
2. IGRP. We have cloned a novel gene that encodes an islet-specific glucose-6-phosphatase catalytic subunit-related protein (IGRP). The goal of the on-going experiments in this project is the elucidation of the molecular basis for the islet-specific expression of the IGRP gene. These experiments involve the use of both tissue culture cells and transgenic mice. The data suggest that the regulation of IGRP gene expression is determined, in part, by novel factors. As such, the identification of these novel factors will potentially aid other investigators who are attempting to understand the process whereby islet stem cells differentiate towards that of a beta cell lineage. In addition, although its biological function is unknown, IGRP has recently been identified as a major autoantigen in type 1 diabetes. Genetic data suggests that suppression of IGRP gene expression might prevent the development of type 1 diabetes in children.
