Research Description
The NF-kappaB family of transcription factors are regulators of many critical processes such as proliferation, apoptosis, immune responses and inflammation. It is, therefore, unsurprising that they play roles in basic development but also in most diseases including cancer. Our laboratory has generated a number of mouse models to investigate these roles in development; breast, lungs, limbs and immune system. We are also interested in the roles played by NF-kappaB signaling in disease. We are currently primarily interested in ovarian, breast and lung cancer. We have generated a number of interesting models that we use for our research. Our transgenic reporter mice express a GFP/luciferase fusion protein that can emit light in response to NF-kappaB activity. With these we can monitor activation in a living animal using a CCD camera and use immunohistochemical techniques to identify specific cell types in which NF-kappaB is active. We have also generated a modular inducible system that enables us to turn up or down NF-kappaB activity in specific cell types eg. mammary or lung epithelium or macrophages. We are using multiple models to study the effects of modulating NF-kappaB activity on normal breast development, early stages of primary breast tumor development and on metastasis to the lungs. We are also collaborating with Professor Todd Giorgio in the Department of Biomedical Engineering to develop nanoparticles that deliver siRNA to macrophages for cancer therapy.

Available Postdoctoral Position Details

We are seeking a postdoctoral fellow to work on a recently funded collaborative project. The studies will include the Yull lab (mouse models and NF-kappaB), the lab of Julie Rhoades (expert in bone biology) and the lab of Todd Giorgio (expert in nanoparticle development and biomedical engineering). The Yull and Giorgio labs have been working together for more than 10 years with co-authored publications, jointly mentored trainees and multiple shared research awards. These are exciting studies that will use multiple methods to modulate Nuclear Factor-kappaB signaling specifically in macrophages to develop new options for treatment of breast metastases in the lungs or bone. Mouse models of metastases will be used to evaluate treatment approaches including doxycycline-inducible transgenic mice, liposomal drugs and or nanoparticle delivery of siRNA to macrophages. Previous experience that would be of value includes; use of mouse models of cancer (particularly in lung and/or bone), flow cytometry, IHC and/or IF, qRT-PCR, cell culture, protein analysis and basic lab skills. Strong communication and social skills and the ability to function with a high level of independence but to mentor junior lab members as needed would be considered a plus. The postdoctoral fellow will participate in lab meetings, research in progress seminars, and presentations at conferences. They will prepare manuscripts based on their research, aid in developing the PI’s grant applications, and will be expected to apply for personal grant funding. The unique collaborative environment provides the opportunity to develop skills and methods that span multiple disciplines. Use of the advanced instrumentation available at Vanderbilt University and the Vanderbilt University Medical Center as shared resources may offer valuable additional skills. Among the key duties of this position are the following: • Designs, performs and analyzes experiments. • Communicates with the PI and presents the data in a timely manner. • Performs specific laboratory techniques that involve animal models of mammary tumor metastasis, qRT-PCR, flow cytometry, in vivo fluorescence imaging, immunohistochemistry/immunofluorescence, protein analysis and cell culture. • Keeps up to date on the relevant literature and assists the PI in preparation of manuscripts and grant applications relevant to own experimental work. • Assists with the instructing and training of students and research assistants in laboratory instrumentation, etc. to develop the skills necessary to perform the required support functions. • Assists in maintaining performance standards, the selection of research methodologies and instrumentation, and coordination of experimental approaches in support of research. • Keeps clear and detailed records of procedures and experiments in accordance with laboratory guidelines, maintains data in shared and readily accessible locations, presents data to the PI and other lab members.
