Research Description
Transient, highly localized calcium signals elicit diverse physiological responses, mediated by calcium sensors, such as calmodulin. Our laboratory studies the regulation and roles of calcium/calmodulin-dependent proteins kinase II (CaMKII), a major target of calcium/calmodulin. CaMKII is highly abundant in the CNS and is highly enriched in neuronal dendritic spines, where it phosphorylates numerous proteins to regulate diverse processes that control the size and function of dendritic spines, leading to learning and memory. Many neuropsychiatric disorders are associated with abnormal numbers, shape or function of dendritic spines. Our goals are to understand the molecular mechanisms that control calcium and CaMKII in dendritic spines, and understand their role in neuropsychiatric diseases.

My lab has pioneered studies of novel CaMKII regulatory mechanisms, and their role in calcium signaling for ~3 decades. We focus on testing an overarching hypothesis that CaMKII activity, substrate specificity, autophosphorylation, subcellular localization, and/or non-enzymatic scaffolding functions are modulated by CaMKII-associated proteins (CaMKAPs), first articulated in 1995 (McNeill, 1995). Over the years, we explored the roles of CaMKII and CaMKAPs in multiple cells/tissues, including the brain, heart, oocytes, and pancreatic beta cells. In the brain, we have studied >10 different CaMKAPs to varying extents, but we recently used proteomics methods to detect >100 proteins that directly or indirectly associate with synaptic CaMKII holoenzymes (Baucum, 2015). Our ongoing efforts are focussed in two main areas. First, we are charactering the biochemical, cellular and behavioral impacts of a de novo E183V mutation in the CaMKIIa catalytic domain that was first identified in an individual diagnosed with autism and later linked to Intellectual Disability (Stephenson, 2017). Second, we are elucidating CaMKII-dependent mechanisms linking neuronal LTCC activation to the stimulation of immediate early gene expression. Initiation of this long-range signal requires direct CaMKII binding to both LTCC a1D subunits (Wang, 2017) and the scaffolding protein, Shank3 (Perfitt, 2020). LTCCs and Shank are frequently mutated in patients with diverse neuropsychiatric disorders, including autism, potentially linking these two projects.


We use a combination of biochemical, molecular, structural, cell imaging, electrophysiological, transgenic mouse and behavioral approaches. The information we obtain could inform the development of novel strategies to treat major human diseases.
